Five cases of papillary thyroid carcinoma (PTC) containing varying amount of leukocyte infiltration was selected. The tissue sections of these five cases were stained with a destain-restain protocol. First, the slides were stained with hematoxylin and eosin (HE) stain and digitized into whole-slide images. Following this, the HE stains were removed and the same tissue sections were stained with a pan-leukocyte CD45 antibody staining and were also digitized. Four of these destain-restained tissue sections were used in the training of a convolutional neural network algorithm and the remaining WSI was used in the testing of the trained algorithm. The trained algorithm was run on all five HE stained WSIs for visual performance evaluation.
